My fiance and I will be having a destination wedding next year. Both of our immediate families will be flying out to take part in the ceremony. We have already asked my sister to be my maid of honor and my fiance has asked my brother to be a groomsman. My fiance wants his brother to be his best man and I would like his sister to stand up with me as well, so both of our siblings could be included. We mentioned asking his brother and sister to stand up to my future mother in law and she is completely against us asking them, saying it would be rude to ask and that there is too much cost involved in standing up. My in parents are paying for my family's accomodations and his parents used their time share to take care of the accomodations for his side. There will be no traveling expense involved for either his brother or sister. Should we still ask his brother and sister to stand up or abide by my mother in law leaving us with only my brother and sister to stand up?

It may be best for your fiance to speak to his mother about this. He could suggest to her that his siblings should decide for themselves. They have every right to decline if they consider it too expensive.
