I have 3 nieces under the age of 6 standing up in my wedding as flower girls, and no boys to stand as my ring bearer. Is it appropriate to have one of my nieces carry the ring pillow?

Sure, this is no problem.
Children tend to become very shy and frightened as they walk down the aisle. With three of them, it could be...well, interesting. It is probably best for them to practice a number of times and get a good night sleep before the wedding.
