I am getting married again and my closest friend already assumes he will be my Best Man. He was my Best man first time around. Is it acceptable to have the same Best man twice? Both my fiance and ex are ok with it.

You say the bride is ok with it but you haven't said if you're OK with this choice. Is there a reason you're hesitating? Is it possible you might not want to have any memories of the past integrated with this new wedding and marriage? If the answer to any of these questions is yes, then take some time to think. If you feel in any way conflicted, explain this to your best friend and select someone new. If you're all right with the choice then, by all means, full speed ahead.
